#StemCells are a wonderful tool to create atrial cardiomyocytes that spontaneously and continuosly beat in a dish… but what happens if we transfect them with the highly potassium selective channelrhodopsin WiChR? #Thread

science.org/doi/full/10.11…
2/ WiChR is a channel that comes from the marine species Wobblia Lunata.

Differently from previosly described K+-conducting channels such as HcKcR1, this channel is characterized by small photocurrent inactivation and improved light sensitivity. #Thread #Physiology #Optogenetics
3/ During my secondment and thanks to the collaboration with @JoH_VieR and other labs, we successfully transfected my hiPSC-derived atrial cardiomyocytes with WiChR.
This beating cluster in the video is one of those that I indented to get sharp electrode recording! #StemCell

An Israeli biotech company, Renewal Bio that recently created mouse embryos using stem cells, has announced plans to make human embryos to harvest tissue for organ transplants and anti-aging procedures.

#Stemcells #biotech #biology #Syntheticbiology #Aging #transplant
The scientist behind the work clarified that:
The synthetic embryos were not "real" embryos and did not have the potential to develop into live animals. But that is partly because they don't have the technology, right now, to do so.
He admits to taking his "ground-breaking" technology to the next level and creating human embryos. The purpose is to harvest tissue to be used in transplant treatments meant to lengthen a person's life and health.

Named Petosemtamab, the antibody MCLA-158 prevents the onset of #metastasis (that is, the spread of #cancer to other vital organs) and slows the growth of primary tumours in experimental models of cancer. Image
The study also lays the groundwork for the use of #organoids in the #DrugDiscovery process undertaken by pharmaceutical companies.

Organoids are patient-derived samples that can be grown in the laboratory, and they reproduce certain aspects of the tumour compartment. Image

Study from @salkinstitute found that partial reprogramming using the #Yamanaka transcription factors (#Oct4, #Sox2, #Klf4 & #cMyc) for short periods of time:

-restored a youthful #epigenetic signature to #aging cells & extends the life span of a #premature #aging mouse model.
-long-term partial reprogramming led to rejuvenating effects in different tissues, such as the #kidney & #skin, and at the #organismal level; duration of the treatment determined the extent of the beneficial effects.
-The rejuvenating effects were associated with a reversion of the #epigenetic clock and metabolic and #transcriptomic changes, including reduced expression of genes involved in the #inflammation, #senescence & #stressresponse pathways.

Important @ICMR statement on evidence based use of #StemCell Therapy based on critical review of literature & claims by large panel of academic clinicians, which I had the privilege to lead. #StemCells often hyped & shouldn't be offered as #StandardOfCare
Inherent public appeal of #StemCellTherapy is being exploited & promoted unethically as a #MagicRemedy to vulnerable patients & should be regulated. There are many diseases where Basic/Translational/Clinical Research can establish its utility @ISSCR @ICMR @fda @ema leads the way!
Acutely aware that patients & their groups want freedom to try whatever might help them & detest when the academic medical community or regulators are so hung up on #RiskBenefitRatio & good evidence and till then say this therapy they cannot take outside regulated clinical trials

1/Short thread on #Longevity
Longevity is a complex challenge, it appears that humans have a rough cap to #lifespan at about 120 years of age (even if you eat healthy, exercise and get lucky with great genes and a great environment).
2/ So the question is how can we fundamentally move past the 120 year cap? #longevity #lifespan
3/ There's promising data around #calorierestriction and #MTOR activating molecules like #rapamycin, which in other species can extend #lifespan by 10-15% (which if we're lucky, for the few it might extend the human lifespan cap to 132-138 years of age). discovermagazine.com/mind/the-two-f…

New paper! We developed an approach to perform scRNA-seq on islets from #stemcells after txp. We found the islets cured #diabetes & matured to very closely resemble real donor islets! Congrats to @WashUBME PhD co-1st authors @punnaug & @KristinaGMaxwel 1/n cell.com/cell-reports/f…
STORY TIME: This project concept has been a fantasy of mine since I was a graduate student, but the technology to do it robustly was simply not there until recently. Even then, we first performed the transplantations for this study in 2018, because of the extended timescales. 2/n
Many labs, including our own, had observed that pancreatic stem cell derivatives (pancreatic progenitors, endocrine progenitors, stem cell-derived islets (SC-islets) at varies stages of maturation) better controlled glycemia with extended time in mice. 3/n

Absolutely delighted to share the lab’s newest study showing that autism-iPSCs diverge from control-iPSCs from early development in a region-specific manner. Heroic work by @neuro_blue & wonderful collaboration with @sbaroncohen biologicalpsychiatryjournal.com/article/S0006-… What did we find? (thread)
Prenatal development may be a key period when differences associated with autism could be established. We studied early neurodevelopment using human iPSCs generated from autistic or typically developing individuals.
We found that autism-iPSCs displayed impaired ability to self-organise into neural rosettes, which are neurogeneic niches that give rise to neural progenitors that subsequently differentiate into neurons.

While our immune system is important for eliminating harmful foreign molecules and/or pathogens (like the virus causing COVID-19), it is equally important that it does not attack our own cells and organs (which is what occurs in autoimmune diseases).
This delicate balance is, in part, maintained by immune-suppressive cells known as T regulatory cells (Tregs).

#BSDBGenSoc2020 These days we all have other things in our minds, it is difficult to concentrate and many of my thoughts R w/ family anf friends in #Spain) but discussing our work, is an important part of our being and it is going to be important in the weeks and months to come.
While many, all, of us rather do al this in person, we are going to have to adjust in the weeks to come. #BSDBGenSoc2020 is a good way to explore how we are going to do it so, THANKS @_BSDB_ and @the_Node for organizing and running this.
My Tweetorial follows the phenomenal one from @TimFulton_1 @SteventonLab (with whom @AMA_Lab work closely) this morning and is very related.

And now onto the tweetorial in the form of a loooong thread.

@NatureMedicine @sloan_kettering @WeillCornell @scell_papers @dana_peer @LabMassague (2/n) human primary #lungcancer is characterized by the emergence of regenerative cell types, typically seen in response to lung injury, but showing striking lineage promiscuity (identity confused!)
@NatureMedicine @sloan_kettering @WeillCornell @scell_papers @dana_peer @LabMassague (3/n) #metastasis exhibits a continuum of more primitive stem to epithelial progenitor states – driven in large part by key embryonic and lung-specifying transcription factors, SOX2 and SOX9.
