Most answers to questions on #rapamycin, including dosing, you may find in this paper, if you read it slowly to its end. Anti-aging doses have no side-effects by definition: highest doses that do not cause side effects and you are comfortable with.

Doses and schedules are individual and must be personalized under doctor supervision. They may range widely. In my case, I never had side effects at relatively high doses (20 mg/2 weeks) compared to typical 6 mg/week. Will shift to 24, probably. But do not do that on your own

For men, body weights 60 kg (132 lb), heights 167 cm; #BMI 21.5.
For women, body weights 52 kg (115 lb); heights 149 cm; BMI 22.8

Low weights are expected of course. But note low heights 167 cm, 149 cm (non-centenarian heights are 179 and 166 for men and women in Austria). One of explanations is that mTOR drives both growth and aging

Of course we cannot (and should not) decrease our heights (developmental growth) but we can decrease mTOR activity later in life by low calories diets, fasting, #keto, #rapamycin etc

These are diverse diseases & increasing age technically not a disease at all. However, there is at least 1 factor in common among all of the co-morbidities identified thus far:

INFLAMMATION

A survey of the co-morbidities revealed another commonality; a close association w/ dysregulation of the pro-inflammatory, neutrophil-recruiting cytokine, IL-17A.

IL-17A is made by multiple immune cell types, including lung-resident innate lymphoid cells (ILC3s).

I predict that so-called more selective mTORC1 inhibitors will have equal or inferior antiaging effects compared with #rapamycin. I predict that maximal anti-aging effect will be achieved with a combination of high doses of rapamycin plus low doses of pan-mTOR inhibitors
such as Torin-1, which can inhibit rapamycin-insensitive functions of #mTORC1. But doses must be low to avoid inhibition of other kinases and side effects. Thus High rapamycin plus Low pan-mTOR inhibitor

"Better" rapalogs are patented #rapalogs so can attract investments and grants. Better rapalogs never showed life extension in any animal yet. They will be better, when, and only when, they extend life longer than "not-better" rapalogs. Better rapalogs were not given to humans.
Even if better, they will not be available soon. Current (“not better”) rapalogs (#rapamycin, everolimus) showed no side effects in all three studies in the elderly. Furthermore, they can be used intermittently and discontinued when any slight doubt emerges.
I always emphasized that no one should take rapamycin or any other rapalogs as self-experimentation, no one. Potential treatment should be performed by a doctor and under doctors’ supervision
