#quantum is here! Why not collect #fluorescence #photon-by-photon in your laser-scanning #microscope? We developed the #BrightEyes_TTM, an #opensource data acquisition system able to do it!

Our time-tagging #DAQ module can collect single-photon signals from 25 independent channels and reference signals from 3 channels. We showed how this module combined with a #SPAD array allows us to perform #FLISM and #FLFS. (2/5)

We characterized the #BrightEyes_TTM. We can reach a single-shot precision of 30 ps, 4 ns dead-time, and an "infinite" range in all channels, with a total flux of up to 125 MHz. (3/5)

Vector-based SARS-CoV2 #vaccines have been associated with vaccine-induced thrombotic thrombocytopenia #VITT/#TTS. It is known that similar to HIT, anti-PF4 antibodies are formed which activate platelets therefore eventually leading to serious thrombosis: nature.com/articles/s4158…
This serious and sometimes fatal condition has been described for both vector-based SARS-CoV2 vaccines ChAdOx1 nCoV-19 (AstraZeneca) and in less frequency for Ad26.COV2.S (Johnson&Johnson). However, it is still unclear why there are large differences in the incidence of #VITT.
To tackle this question, we dropped everything else and compared both vaccines in our local, highly interdisciplinary multi-expertise group from @uni_greifswald, @UMGreifswald and @Loeffler_News: #GreifswalderForscher

1/9 Could graphene coverslips improve your single-molecule fluorescence and superresolution experiments? In our article in @AdvancedAms we combine #DNAorigami with #graphene to perform #singlemolecule #biophysics, #biosensing & #superresolution microscopy.
2/9 Graphene is almost transparent and quenches fluorescence in a range of ~40 nm. Fluorescence intensity or fluorescence lifetime of single molecules directly tell you the distance of a molecule to the surface. Differences in the z-direction as small as 2.9 nm are resolved. Image
3/9 Dynamics with graphene energy transfer (GET). We use GET to visualize motion on top of a graphene surface and determine the timescale of fluctuations via autocorrelation analysis. Image

We are particularly interested in characterizing the limits of #deeplearning based approaches to image restoration. You will find several experiments comparing several best-in-class approaches: #3DRCAN, #CARE, #SRResNet and #ESRGAN.

(1/6) Excited to share that our OligoFISSEQ paper is up (nature.com/articles/s4159…)! We set out to develop single cell in situ #3Dgenome mapping methods. A fun collaborative effort led by @huy_sauce @ShyamtanuC and David Castillo, from @TingWu_Lab and @mamartirenom groups.
(2/6) An idea that began with @SonCNguyen and Evan Daugharthy. The rest of the team including: @GuyNir5 @AntoniosLioutas @ElliotHershberg @Nuno_MCMartins @PaulReginato Mohammed Hannan @oligopain and @geochurch . Shout out to everyone @naturemethods for the great experience!
(3/6) To target and visualize genomic regions at genome-wide scale, we barcoded Oligopaints, increasing target capacity exponentially. We then visualize and map the targets by #InSituSequencing the barcodes using hybridization, synthesis, and ligation-based approaches.

#Pumpy paper is out @NatureComms - "Automating Multimodal Microscopy with NanoJ-Fluidics"! We developed an open-source inexpensive #LEGO-based fluidics system that can be coupled to a microscope to achieve high-performance imaging nature.com/articles/s4146…
We created video abstract with @LaineBioImaging and @christlet that quickly explains paper. #Pumpy enables live-to-fixed imaging, fixation-on-event, super-resolution sequential-labelling (unlim. number protein species), it adds a new dimension to what’s possible at the microscope
@LaineBioImaging @christlet @palmada86 @P_Matos_Pereira did an excellent job making #Pumpy happen. An excellent collaboration with the Charras, @BaumBuzz and @christlet labs. Kudos to @christlet, despite the distance we interacted almost on a daily basis getting the paper together
